I'd say it would have to be very large, considering that wolves in the wild hold territories that encompass miles of land. Depends on how many wolves really. The more wolves, the more territory needed to feed them all. Wolves in Alberta apparently have territories encompassing 357-1779 square km of land, so I doubt any sanctuary could manage that.

Wolves are intelligent animals and bore easily. Novelty objects introduced every few days or so (chew toys, new flavour meats etc.) would keep their minds stimulated.
There is now a growing band of us, who came to the African bush with all our prejudices, with all that 'common knowledge' about hyenas which proved so totally wrong, and who just fell for the spell of animals which were so totally different- Hans Kruuk

Captive Wolves enclosures range from tiny Zoo enclosures to Acres of fenced in land like Haliburton Forest Wolf Center which has 15 acres.(One of the Largest in the world)

Enrichment activity for captive animals of intelligence is important to their health & well being.
At Wolf Howl Animal Preserve, they are constantly trying new things to keep the Wolves entertained 8)

Wooden & Rock Platforms in the enclosure give the wolves higher vantage points. Toys, like "Kongs" filled with treats, Pumpkins at Halloween, Eggs at Easter, A Sausage decorated Tree at Christmas

It depends on how many wolves, and the setting.

I am just assuming you are looking to make an enclosure, and not designating a large area of land as an open sanctuary.

As a general rule, you want a minimum of 2 acres for 2 to 4 wolves.

Climate is also very important to consider. Some form of shade is essential, so trees or man made shelters are a good idea.

Wolf cubs like toys, but adults generally amuse themselves by playing with the other wolves in their enclosure. You will want to have some large rocks, and other objects which they can hide behind and ambush each other as they play.

There is a lot to consider when you are putting something like this together. If you would like any suggestions, I would be more than happy to talk to you about it. Feel free to PM me.

Haliburton wolf center is 15 acres, which seems like a fair size for captive wolves.
